PURPOSE: Chagas disease (CD) a Neglected Tropical Diseases is an important public health issue in countries where is still endemic, included in the Sustainable Development Goals (SDG). Traditionally restricted to rural areas with diverse routes of transmissions from vectorial to oral with acute manifestations but being more common diagnosed in chronic stages. The aim of this investigation was to characterize the Knowledge, Attitudes and Practices (KAP) related to Chagas disease (CD) in two rural settlements of the Colombian Caribbean with previous records of the disease and/or the parasite. METHODS: A cross-sectional descriptive study was made in two rural settlements in Colombia and surveillance instrument was developed to measure Knowledge, Attitudes and Practices (KAP) related to Chagas disease (CD). RESULTS: In a population with > 60% women and access to social security around 66.5%; 81,6% were homeowners with access to water and electricity > 90% but only 9% of sewerage. The level of knowledge about CD was around 62% but lack of specificity about comprehension of transmission routes (74,6%), and symptoms (85,3%) were found; concluding that 86% of the surveyed sample had very poor level of knowledge about the disease despite preventive campaigns carried out in the two communities studied. CONCLUSIONS: Despite of a low frequency of CD in this Caribbean areas, the presence of vector, risk factors plus poor level of knowledge about the disease justify that public health intervention strategies should be implemented and monitored over time to maintain uninterrupted surveillance of Chagas Disease.

The co-culturing of microorganisms is a well-known strategy to study microbial interactions in the laboratory. This approach facilitates the identification of new signals and molecules produced by one species that affects other species' behavior. In this work, we have studied the effects of the interaction of nine Streptomyces species (S. albidoflavus, S. ambofaciens, S. argillaceus, S. griseus, S. lividans, S. olivaceus, S. parvulus, S. peucetius, and S. rochei) with the predator bacteria Myxococcus xanthus, five of which (S. albidoflavus, S. griseus, S. lividans, S. olivaceus, and S. argillaceus) induce mound formation of M. xanthus on complex media (Casitone Yeast extract (CYE) and Casitone tris (CTT); media on which M. xanthus does not form these aggregates under normal culture conditions. An in-depth study on S. griseus-M. xanthus interactions (the Streptomyces strain producing the strongest effect) has allowed the identification of two siderophores produced by S. griseus, demethylenenocardamine and nocardamine, responsible for this grouping effect over M. xanthus. Experiments using pure commercial nocardamine and different concentrations of FeSO4 show that iron depletion is responsible for the behavior of M. xanthus. Additionally, it was found that molecules, smaller than 3 kDa, produced by S. peucetius can induce the production of DK-xanthenes by M. xanthus.

Introducción: La enfermedad hepática grasa no alcohólica es una afección clínica- morfológica que se caracteriza por una infiltración grasa del hígado en más de un 5 %; tiene dos estadios: esteatosis simple y esteatohepatitis, la cual puede progresar a: fibrosis, cirrosis y hepatocarcinoma. Objetivo: Determinar la relación entre las variables clínicas y epidemiológicas con esta enfermedad, así como los índices de fibrosis y su relación. Métodos: Se realizó un estudio descriptivo con diseño transversal en los pacientes atendidos en la consulta de hígado y vías biliares del Hospital Universitario « Dr. Celestino Hernández Robau», en la etapa de enero- diciembre de 2017. Se trabajó con una población conformada por 60 pacientes, mayores o igual a 20 años de edad, con diagnóstico de enfermedad hepática grasa no alcohólica primaria, sobrepesos u obesos. Resultados: Predominó el grupo etario entre 50-59 años de edad, del sexo femenino, obesos y con esteatosis grado I. Se constató que el 73,33% de los pacientes tenían síndrome metabólico y en ellos prevaleció el grado II de esteatosis. Al relacionar los riesgos de fibrosis se encontraron 28 pacientes con riesgo indeterminado y alto en las clasificaciones FIB-4 y NFS, respectivamente, y 5 presentaron alto riesgo en ambas variables. Los índices de FIB-4 y NFS tuvieron una correlación significativa, directamente proporcional y considerable. Conclusiones: La correlación detectada entre los índices FIB-4 y NFS reafirmó el valor en la detección de sospecha de fibrosis y orientó, en la práctica clínica, la conducta ante los diferentes pacientes con esta afección.


Introduction: non-alcoholic fatty liver disease is a clinical and morphological condition characterized by fatty infiltration of the liver in more than 5%; it has two stages: simple steatosis and steatohepatitis, which can progress to fibrosis, cirrhosis and hepatocellular carcinoma. Objective: to determine the relationship between clinical and epidemiological variables with this disease, as well as fibrosis indices and their relationship. Methods: a cross-sectional descriptive study was carried out in patients seen in the liver and biliary tract consultation at " Dr. Celestino Hernández Robau" University Hospital from January to December 2017. We worked with a population made up of 60 overweight or obese patients older than or equal to 20 years who were diagnosed with primary non-alcoholic fatty liver disease. Results: the age group between 50-59 years of age, female gender, obese ones and with grade I steatosis prevailed. We found that 73.33% of the patients had metabolic syndrome and grade II steatosis prevailed in them. A number of 28 patients were found with indeterminate and high risk in the FIB-4 and NAFLD classifications, respectively, when relating the risks of fibrosis, as well as 5 had high risk in both variables. The FIB-4 and NAFLD indices had a significant, directly proportional, and considerable correlation. Conclusions: the correlation detected between the FIB-4 and NAFLD indices reaffirmed the value in the detection of suspected fibrosis and guided, in clinical practice, the conduct of different patients with this condition.

Bacteria of the genus Streptomyces have a plethora of transcriptional regulators, among which the xenobiotic response element (XRE) plays an important role. In this organism, XRE regulators are often followed downstream by small proteins of unknown function containing a DUF397 domain. It has been proposed that XRE/DUF397 pairs constitute type II toxin-antitoxin (TA) systems. However, previous work carried out by our group has shown that one of these systems is a strong activator of antibiotic production in S. coelicolor and other Streptomyces species. In this work, we have studied the overexpression of fourteen XRE/DUF397 pairs present in the S. coelicolor genome and found that none behave as a type II TA system. Instead, they act as pleiotropic regulators affecting, in a dependent manner, antibiotic production and morphological differentiation on different culture media. After deleting, individually, six XRE/DUF397 pairs (those systems producing more notable phenotypic changes when overexpressed: SCO2246/45, SCO2253/52, SCO4176/77, SCO4678/79, SCO6236/35, and SCO7615/16), the pair SCO7615/16 was identified as producing the most dramatic differences as compared to the wild-type strain. The SCO7615/16 mutant had a different phenotype on each of the media tested (R2YE, LB, NMMP, YEPD, and MSA). In particular, on R2YE and YEPD media, a bald phenotype was observed even after 7 days, with little or no actinorhodin (ACT) production. Lower ACT production was also observed on LB medium, but the bacteria were able to produce aerial mycelium. On NMMP medium, the mutant produced a larger amount of ACT as compared with the wild-type strain.

Bacteria of the Streptomyces genus constitute an authentic biotech gold mine thanks to their ability to produce a myriad of compounds and enzymes of great interest at various clinical, agricultural, and industrial levels. Understanding the physiology of these organisms and revealing their regulatory mechanisms is essential for their manipulation and application. Two-component systems (TCSs) constitute the predominant signal transduction mechanism in prokaryotes, and can detect a multitude of external and internal stimuli and trigger the appropriate cellular responses for adapting to diverse environmental conditions. These global regulatory systems usually coordinate various biological processes for the maintenance of homeostasis and proper cell function. Here, we review the multiple TCSs described and characterized in Streptomyces coelicolor, one of the most studied and important model species within this bacterial group. TCSs are involved in all cellular processes; hence, unravelling the complex regulatory network they form is essential for their potential biotechnological application.

Streptomyces bacteria produce a wide number of antibiotics and antitumor compounds that have attracted the attention of pharmaceutical and biotech companies. In this study, we provide evidence showing that the xylem sap from grapevines has a positive effect on the production of different antibiotics by several Streptomyces species, including S. ambofaciens ATCC 23877 and S. argillaceus ATCC 12596 among others. The production of several already known compounds was induced: actinomycin D, chromomycin A3, fungichromin B, mithramycin A, etc., and four compounds with molecular formulas not included in the Dictionary of Natural Products (DNP v28.2) were also produced. The molecules present in the xylem sap that acts as elicitors were smaller than 3 kDa and soluble in water and insoluble in ether, ethyl acetate, or methanol. A combination of potassium citrate and di-D-fructose dianhydrides (related to levanbiose or inulobiose) seemed to be the main effectors identified from the active fraction. However, the level of induction obtained in the presence of these compounds mix was weaker and delayed with respect to the one got when using the whole xylem sap or the 3 kDa sap fraction, suggesting that another, not identified, elicitor must be also implied in this induction.

Introducción: La pérdida del anillo neurorretiniano (ANR) es una característica distintiva del glaucoma que genera desviación de la regla ISNT. Objetivos: Determinar el porcentaje de ojos sanos que cumplen la regla ISNT y establecer cuál de sus variantes es más aplicable a la población normal. Métodos: Evaluación del orden del grosor del anillo neurorretiniano por medio de fotografía de disco óptico, capa de fibras nerviosas en tomografía de coherencia óptica (OCT) y evaluación clínica por oftalmoscopia indirecta. Resultados: Se evaluaron 102 ojos sanos de pacientes con edad promedio 44.1 años. El porcentaje de la regla ISNT en foto de disco, OCT y oftalmoscopia indirecta se cumplió en el 36.3, 38.2 y 29.4% de los ojos respectivamente, la variación de la regla en la que se excluye cuadrante nasal y temporal, conocida como regla «IS¼, aumentó sus porcentajes de cumplimiento al 73.5, 52.9 y 54.9% al ser evaluadas en foto de disco, OCT y oftalmoscopia indirecta, respectivamente. Conclusiones: La regla ISNT solo es válida en un tercio de los ojos evaluados mediante los métodos descrito


Background: Neurorretinal rim loss is an important characteristic of glaucoma that generates deviation from the ISNT rule. Objective: To determine what percentage of normal eyes follow the ISNT rule, and wich ISNT rule variants may be more generalizable to the normal population. Methods: Neurorretinal rim assessment from optic disc photographs, retinal nerve fiber layer (RNFL) thickness measurements from (OCT) and clinical evaluation by indirect ophthalmoscopy. Results: 102 healthy eyes were evaluated, with an average age of 44.1 ± 11.7 years. The percentage of agreement of ISNT rule was verified in in disk photo (36.3%), OCT (38.2%) and indirect ophthalmoscopy (29.4%) of the total population. The variation of the rule in which the nasal (73.5) and temporal (52.9%) quadrant are excluded, known as the "IS" rule, increased its compliance percentages from 73.5%, 52.9% and 54.9% when evaluated in disk photo, OCT and indirect ophthalmoscopy, respectively.

Ocean currents, multiple fecal bacteria input sources, and jurisdictional boundaries can complicate pollution source tracking and associated mitigation and management efforts within the nearshore coastal environment. In this study, multiple microbial source tracking tools were employed to characterize the impact and reach of an ocean wastewater treatment facility discharge in Mexico northward along the coast and across the Southwest United States- Mexico Border. Water samples were evaluated for fecal indicator bacteria (FIB), Enterococcus by culture-based methods, and human-associated genetic marker (HF183) and Enterococcus by droplet digital polymerase chain reaction (ddPCR). In addition, 16S rRNA gene sequence analysis was performed and the SourceTracker algorithm was used to characterize the bacterial community of the wastewater treatment plume and its contribution to beach waters. Sampling dates were chosen based on ocean conditions associated with northern currents. Evidence of a gradient in human fecal pollution that extended north from the wastewater discharge across the United States/Mexico border from the point source was observed using human-associated genetic markers and microbial community analysis. The spatial extent of fecal contamination observed was largely dependent on swell and ocean conditions. These findings demonstrate the utility of a combination of molecular tools for understanding and tracking specific pollutant sources in dynamic coastal water environments.

We describe an Oropouche orthobunyavirus infection in a women 28 years of age in Colombia. We confirmed the diagnosis by viral isolation, quantitative reverse transcription PCR, and phylogenetic analysis of the small, medium, and large genomic segments. The virus is related to a strain isolated in Ecuador in 2016.

RESUMEN Introducción: Los avances tecnológicos en tomografía computarizada han hecho posible la evaluación de estructuras pequeñas y móviles, como las arterias coronarias y el resto del corazón. La tomografía cardíaca aporta información anatómica y funcional. Objetivo: Identificar los valores de las principales medidas anatómicas de las cavidades cardíacas y los grandes vasos, y compararlos con los valores de referencia utilizados. Método: Se realizó un estudio analítico de investigación y desarrollo con un universo de 325 pacientes normotensos, no obesos, sin enfermedad estructural cardiopulmonar o de grandes vasos, arritmia o enfermedad coronaria significativa, de donde se seleccionaron 100 pacientes por muestreo probabilístico aleatorio simple, a quienes se les realizó tomografía computarizada cardíaca de doble fuente en el Cardiocentro Ernesto Che Guevara, Santa Clara, Cuba. Resultados: Las mediciones ventriculares izquierda y derecha mostraron diferencias con la mayoría de los valores referenciales. Los diámetros de la aorta, aumentaron con la edad, fueron superiores en los hombres y difieren de los valores de referencia con excepción del medido en eje corto de la raíz aórtica en telediástole, y el diámetro en eje corto de la aorta descendente en telesístole. La arteria pulmonar mostró diferencias significativas con relación a los valores de referencia. Conclusiones: Las mediciones de las cavidades cardíacas y los grandes vasos basados en las técnicas tomográficas constituyen una necesidad en determinados grupos poblacionales para lograr una estandarización adecuada debido a su gran variabilidad en relación a distintas variables. La diferencia significativa con los valores de referencia indica le necesidad de estudios multicéntricos con poblaciones mayores para lograr su homogeneidad.


ABSTRACT Introduction: Technological advances in computed tomography have made it possible to assess small, mobile structures such as the coronary arteries and the rest of the heart. Cardiac tomography provides anatomical and functional information. Objective: To identify the values of the main anatomical sizes of the cardiac chambers and great vessels, and compare them with the reference values. Method: An analytical research and development study was conducted with a population of 325 normotensive, non-obese patients without cardiopulmonary or great vessel structural disease, arrhythmia or significant coronary artery disease. One hundred patients were selected by simple random probability sampling, who underwent dual-source cardiac computed tomography at the Cardiocentro Ernesto Che Guevara, Santa Clara, Cuba. Results: Left and right ventricular measurements showed differences when compared with most reference values. Aortic diameters increased with age. They were higher in men and were different from the reference values except for the short-axis diameter of the aortic root in end-diastole and short-axis diameter of the descending aorta in end-systole. The pulmonary artery showed significant differences in relation to the reference values. Conclusions: Measurements of cardiac chambers and great vessels by tomographic techniques are necessary in certain population groups to achieve adequate standardization due to their great variability in relation to different variables. The significant difference with the reference values indicates the need for multicenter studies with larger populations to achieve homogeneity.

This report is concerned with the relevance of the microscopic rules that implement individual neuronal activation, in determining the collective dynamics, under variations of the network topology. To fix ideas we study the dynamics of two cellular automaton models, commonly used, rather in-distinctively, as the building blocks of large-scale neuronal networks. One model, due to Greenberg and Hastings (GH), can be described by evolution equations mimicking an integrate-and-fire process, while the other model, due to Kinouchi and Copelli (KC), represents an abstract branching process, where a single active neuron activates a given number of postsynaptic neurons according to a prescribed "activity" branching ratio. Despite the apparent similarity between the local neuronal dynamics of the two models, it is shown that they exhibit very different collective dynamics as a function of the network topology. The GH model shows qualitatively different dynamical regimes as the network topology is varied, including transients to a ground (inactive) state, continuous and discontinuous dynamical phase transitions. In contrast, the KC model only exhibits a continuous phase transition, independently of the network topology. These results highlight the importance of paying attention to the microscopic rules chosen to model the interneuronal interactions in large-scale numerical simulations, in particular when the network topology is far from a mean-field description. One such case is the extensive work being done in the context of the Human Connectome, where a wide variety of types of models are being used to understand the brain collective dynamics.

PURPOSE: The aim of the present work was to assess the electrogenic activity of bacteria from hydrothermal vent sediments achieved under sulfate reducing (SR) conditions in a microbial fuel cell design with acetate, propionate and butyrate as electron donors. METHODS: Two different mixtures of volatile fatty acids (VFA) were evaluated as the carbon source at two chemical oxygen demand (COD) proportions. The mixtures of VFA used were: acetate, propionate and butyrate COD: 3:0.5:0.5 (stage 1) and acetate - butyrate COD: 3.5:0.5 (stage 2). Periodical analysis of sulfate (SO4 -2), sulfide (HS-) and COD were conducted to assess sulfate reduction (SR) and COD removal along with measurements of voltage and current to assess the global performance of the consortium in the system. RESULTS: Percentage of SR was of 97.5 ± 0.7 and 74.3 ± 1.5% for stage 1 and 2, respectively. The % COD removal was of 91 ± 2.1 and 75.3 ± 9.6 for stage 1 and 2, respectively. Although SR and COD removal were higher at stage 1, in regards of energy, stage 2 presented higher current and power densities and Coulombic efficiency as follows: 741.7 ± 30.5 µA/m2, 376 ± 34.4 µW/m2 and 5 ± 2.7%, whereas for stage 1 these values were: 419 ± 71 µA/m2, 52.7 ± 18 µW/m2 and 0.02%, respectively. A metagenomic analysis - stage 2 - in the anodic chamber, demonstrated that SR was due to Dethiosulfovibrionaceae (HA73), Desulfobacter and Desulfococcus and the electrogenic microorganisms were Planococcus, SHD-231, Proteiniclasticum, vadinCA02, and families Porphyromonadacea and Pseudomonadaceae. CONCLUSIONS: It was demonstrated that microorganisms prevenient from hydrothermal vent sediments adapted to a microbial fuel cell system are able to generate electricity coupled to 74.3 ± 1.5 and 75.3 ± 9.6% of SR and COD removal respectively, with a mixture of acetate - butyrate.

Actinomycetes constitute a large group of Gram-positive bacteria present in different habitats. One of these habitats involves the association of these bacteria with insects. In this work, we have studied twenty-four actinomycetes strains isolated from the intestinal tract and feces from larvae of the xylophagous coleopteran Cerambyx welensii and have shown that seventeen strains present hydrolytic activity of some of the following substrates: cellulose, hemicellulose, starch and proteins. Fourteen of the isolates produce antimicrobial molecules against the Gram-positive bacteria Micrococcus luteus. Analysis of seven strains led us to identify the production of a wide number of compounds including streptanoate, alpiniamide A, alteramides A and B, coproporphyrin III, deferoxamine, demethylenenocardamine, dihydropicromycin, nocardamine, picromycin, surugamides A, B, C, D and E, tirandamycins A and B, and valinomycin. A significant number of other compounds, whose molecular formulae are not included in the Dictionary of Natural Products (DNP), were also present in the extracts analyzed, which opens up the possibility of identifying new active antibiotics. Molecular identification of ten of the isolated bacteria determined that six of them belong to the genus Streptomyces, two of them are included in the genus Amycolatopsis and two in the genus Nocardiopsis.

Antibiotic resistance currently presents one of the biggest threats to humans. The development and implementation of strategies against the spread of superbugs is a priority for public health. In addition to raising social awareness, approaches such as the discovery of new antibiotic molecules and the elucidation of resistance mechanisms are common measures. Accordingly, the two-component system (TCS) of Streptomyces coelicolor AbrB1/B2, offer amenable ways to study both antibiotic production and resistance. Global transcriptomic comparisons between the wild-type strain S. coelicolor M145 and the mutant ΔabrB, using RNA-Seq, showed that the AbrB1/B2 TCS is implicated in the regulation of different biological processes associated with stress responses, primary and secondary metabolism, and development and differentiation. The ΔabrB mutant showed the up-regulation of antibiotic biosynthetic gene clusters and the down-regulation of the vancomycin resistance gene cluster, according to the phenotypic observations of increased antibiotic production of actinorhodin and undecylprodigiosin, and greater susceptibility to vancomycin. The role of AbrB1/B2 in vancomycin resistance has also been shown by an in silico analysis, which strongly indicates that AbrB1/B2 is a homolog of VraR/S from Staphylococcus aureus and LiaR/S from Enterococcus faecium/Enterococcus faecalis, both of which are implied in vancomycin resistance in these pathogenic organisms that present a serious threat to public health. The results obtained are interesting from a biotechnological perspective since, on one hand, this TCS is a negative regulator of antibiotic production and its high degree of conservation throughout Streptomyces spp. makes it a valuable tool for improving antibiotic production and the discovery of cryptic metabolites with antibiotic action. On the other hand, AbrB1/B2 contributes to vancomycin resistance and is a homolog of VraR/S and LiaR/S, important regulators in clinically relevant antibiotic-resistant bacteria. Therefore, the study of AbrB1/B2 could provide new insight into the mechanism of this type of resistance.

: The prevalence of hypertension, type 2 diabetes mellitus (DM2) and the metabolic syndrome continues to increase in Latin America, while the rates of diagnosis, treatment and control of these disorders remain low. The frequency of the risk factors that constitute the metabolic syndrome and are associated with an increased risk of cardiovascular disease has not diminished since the publication of the previous consensus. This document discusses the socioeconomic, demographic, environmental and cultural characteristics of most associated Latin American countries and partially explains the lack of better results in improving clinical and public health actions that allow high morbidity and mortality rates caused by cardiovascular diseases and DM2 to be reduced through programs aligned with the so-called precision medicine, which should be predictive, preventive, personalized and participatory. The Consensus ratifies the diagnostic criteria expressed in the previous consensus to define hypertension and DM2 but, for the metabolic syndrome, and in the absence of evidence, the recommendation is to implement a cohort study that determines the abdominal perimeter value associated with hard outcomes, such as DM2 and CVD. Meanwhile, we recommend modifying the criterion to more than 94 cm in men and more than 84 cm in women according to WHO recommendations. We also recommend the carrying out of a study that identifies the situation of hypertension and DM2 in people of African ancestry who, in Latin America, exceed 75 million and whose epidemiology does not include solid studies. With respect to the proposed therapeutic targets, we recommended maintaining those defined in the previous consensus, but insisting that early pharmacological management of prediabetes with metformin should be introduced, as should the treatment of diabetic hypertensive patients with a combination therapy of two fixed-dose antihypertensive drugs and management with statins. To increase adherence, the use of different drugs combined in a single pill (polypill) is recommended. The simplification of the therapeutic regimen is accompanied by greater control of cardiovascular risk factors, both in primary and secondary prevention, and has been shown to be cost-effective. The consensus recommends the use of the currently available polypill combining an angiotensin-converting enzyme inhibitor, a statin and aspirin for secondary cardiovascular prevention and in patients with a high cardiovascular risk, such as hypertension patients with DM2.

The xenobiotic response element (XRE) transcription factors belong to a regulator family frequently found in Streptomyces that are often followed by small proteins with a DUF397 domain. In fact, the pair XRE-DUF397 has been proposed to comprise toxin-antitoxin (TA) type II systems. In this work, we demonstrate that one of these putative TA-systems, encoded by the genes SCO4441 and SCO4442 of Streptomyces coelicolor, and denominated Scr1/Scr2 (which stands for S. c oelicolor r egulator), does not behave as a toxin-antitoxin system under the conditions used as was originally expected. Instead the pair Scr1/Scr2 acts as a strong positive regulator of endogenous antibiotic production in S. coelicolor. The analysis of the 19 Streptomyces strains tested determined that overexpression of the pair Scr1/Scr2 drastically induces the production of antibiotics not only in S. coelicolor, but also in Streptomyces lividans, Streptomyces peucetius, Streptomyces steffisburgensis and Streptomyces sp. CA-240608. Our work also shows that Scr1 needs Scr2 to exert positive regulation on antibiotic production.

The present study focuses on the development and optimization of a packed-bed reactor (PBR) for continuous production of xylooligosaccharides (XOS) from xylan. For this purpose, three different methacrylic polymer-based supports (Relizyme R403/S, Purolite P8204F and Purolite P8215F) activated with glyoxyl groups were morphologically characterized and screened for the multipoint covalent immobilization of a xylanase. Based on its physical and mechanical properties, maximum protein loading and thermal stability, Relizyme R403/S was selected to set up a PRB for continuous production of XOS from corncob xylan. The specific productivity for XOS at 10 mL/min flow rate was 3277 gXOS genzyme-1 h-1 with a PBR. This PBR conserved >90% of its initial activity after 120 h of continuous operation.

Sequencing of Streptomyces genomes has revealed they harbor a high number of biosynthesis gene cluster (BGC), which uncovered their enormous potentiality to encode specialized metabolites. However, these metabolites are not usually produced under standard laboratory conditions. In this manuscript we report the activation of BGCs for antimycins, carotenoids, germicidins and desferrioxamine compounds in Streptomyces argillaceus, and the identification of the encoded compounds. This was achieved by following different strategies, including changing the growth conditions, heterologous expression of the cluster and inactivating the adpAa or overexpressing the abrC3 global regulatory genes. In addition, three new carotenoid compounds have been identified.

Mayaro virus (MAYV), Venezuelan equine encephalitis virus (VEEV), and chikungunya virus (CHIKV) are vector-borne alphaviruses that cocirculate in South America. Human infections by these viruses are frequently underdiagnosed or misdiagnosed, especially in areas with high dengue virus endemicity. Disease may progress to debilitating arthralgia (MAYV, CHIKV), encephalitis (VEEV), and death. Few standardized serological assays exist for specific human alphavirus infection detection, and antigen cross-reactivity can be problematic. Therefore, serological platforms that aid in the specific detection of multiple alphavirus infections will greatly expand disease surveillance for these emerging infections. In this study, serum samples from South American patients with PCR- and/or isolation-confirmed infections caused by MAYV, VEEV, and CHIKV were examined by using a protein microarray assembled with recombinant capsid, envelope protein 1 (E1), and E2 from nine New and Old World alphaviruses. Notably, specific antibody recognition of E1 was observed only with MAYV infections, whereas E2 was specifically targeted by antibodies from all of the alphavirus infections investigated, with evidence of cross-reactivity to E2 of o'nyong-nyong virus only in CHIKV-infected patient serum samples. Our findings suggest that alphavirus structural protein microarrays can distinguish infections caused by MAYV, VEEV, and CHIKV and that this multiplexed serological platform could be useful for high-throughput disease surveillance. IMPORTANCE Mayaro, chikungunya, and Venezuelan equine encephalitis viruses are closely related alphaviruses that are spread by mosquitos, causing diseases that produce similar influenza-like symptoms or more severe illnesses. Moreover, alphavirus infection symptoms can be similar to those of dengue or Zika disease, leading to underreporting of cases and potential misdiagnoses. New methods that can be used to detect antibody responses to multiple alphaviruses within the same assay would greatly aid disease surveillance efforts. However, possible antibody cross-reactivity between viruses can reduce the quality of laboratory results. Our results demonstrate that antibody responses to multiple alphaviruses can be specifically quantified within the same assay by using selected recombinant protein antigens and further show that Mayaro virus infections result in unique responses to viral envelope proteins.

Bacterial genotyping methods have been used in several areas of microbiology and have facilitated the identification of bacterial strains, as well as the study of virulence and resistance factors, epidemiological surveillance, among others. Constituting, in this way, as complementary or alternative to phenotypic methods. This chapter reviews some of the methods of bacterial genotyping used nowadays, explains briefly how they are performed, and highlights some limitations and advantages they have.
